چکیده

Quantitative light absorption data of the neutral free flavosemiquinone are reported. It is shown that the visible range of the spectrum of the radical is drastically dependent on the solvent. The spectra of the neutral blue flavosemiquinone are compared with those of the blue flavoprotein radicals and conclusions are drawn concerning the environment of the protein-bound flavosemiquinone. The stabilization of the neutral blue flavosemiquinone through N(5)-alkylation and/or n-complex formation with aromatic compounds is described. The light absorption characteristics of a tautomeric neutral, but red-coloured, flavosemiquinone are described. The results are compared with those obtained from the free anionic and protein-bound red flavosemiquinones. The two types of radicals cannot be distinguished from each other by electron spin resonance spectrometry but may be recognized by means of light absorption spectrophotometry. The possible biological significance of the neutral red flavosemiquinone is discussed. The optical properties of this radical are compared with those of free and protein-bound anionic flavosemiquinones.

A flavin binding cryptochrome photoreceptor responds to both blue and red light in Chlamydomonas reinhardtii.

Cryptochromes are flavoproteins that act as sensory blue light receptors in insects, plants, fungi, and bacteria. We have investigated a cryptochrome from the green alga Chlamydomonas reinhardtii with sequence homology to animal cryptochromes and (6-4) photolyases. In response to blue and red light exposure, this animal-like cryptochrome (aCRY) alters the light-dependent expression of various g...

Light-triggered proton and electron transfer in flavin cofactors.

The pH dependent behavior of two flavin cofactors, flavin-adenine dinucleotide (FAD) and flavin mononucleotide (FMN), has been characterized using femtosecond transient absorption spectroscopy for the first time. The flavin excited state was characterized in three states of protonation (Fl(-), Fl, and FlH(+)). We found that Fl and Fl(-) exhibit the same excited state absorption but that the lif...

On the interaction of flavins with phosphine-derivatives.

The interaction of flavinium salts with triphenylphosphine is described. The complexes which can be isolated in high yields in the crystalline state contain one mole triphenylphosphine per mole flavin. Triphenylphosphine is covalently bound to the N (5)-atom of the isoalloxazine ring system. Dissociation constants and rate constants have been determined spectrophotometrically. Evidence for the ...

Role of Charge-transfer Interactions in Flavoprotein Catalysis

The last decade has produced a veritable explosion of knowledge on the chemical reactivity of the isoalloxazine ring system of the flavin coenzymes, largely because of a side-by-side development with knowledge of flavoenzymes and model flavin studies. In this period the visible absorption spectra and epr spectra of the flavin semiquinone in its cationic, neutral, and anionic forms have been def...

Cryptochrome blue light photoreceptors are activated through interconversion of flavin redox states.

Cryptochromes are blue light-sensing photoreceptors found in plants, animals, and humans. They are known to play key roles in the regulation of the circadian clock and in development. However, despite striking structural similarities to photolyase DNA repair enzymes, cryptochromes do not repair double-stranded DNA, and their mechanism of action is unknown. Recently, a blue light-dependent intra...
